Acetoin: Polarographic Determination and Disappearance from the Blood after Administration

ثبت نشده
چکیده

Acetoin (acetylmethylcarbinol) has been found in minute amounts in normal human urine (1) and in the blood of the cow, pig, sheep, and horse (2, 3). Westerfeld, Stotz, and Berg (4) have reported that in dogs the rate of disappearance of alcohol from the blood was increased by the administration of pyruvate and that of pyruvate by the administration of alcohol. They interpret their results as indicating that acetaldehyde, produced by a primary oxidation of alcohol, may be condensed with pyruvate to form acetoin. They suggest that this reaction may furnish an explanation for a relationship between the metabolism of alcohol and of carbohydrate. Support is given to their suggestion by the fact that pyruvic acid is a product of the normal metabolism of dextrose and that the condensation of acetaldehyde and pyruvic acid to form acetoin is known to occur in the metabolic activity of yeast and bacteria. Green, Westerfeld, Vennesland, and Knox (5) have demonstrated this condensation in vitro with enzymes from tissues of various animals. Westerfeld, Stotz, and Berg did not test their interpretation of alcohol metabolism by making determinations for acetoin in the blood of the dogs to which they gave alcohol and pyruvate. The investigation reported here is preliminary to such determination (6). The disappearance of acetoin from the blood following its administration was studied, since the possibility exists that it may be so rapid as to prevent the accumulation of appreciable amounts of acetoin.

A colorimetric determination of blood acetoin.

Most of the methods described for the determination of acetoin and biacetyl are based on the original Lemoigne-van Niel procedure (1, 2), which depends upon the formation of insoluble nickel dimethylglyoxime when biacetyl is treated with hydroxylamine in the presence of a nickel salt; acetoin is similarly determined after oxidation to biacetyl by heating with ferric chloride. In a recent modifi...

Differential Pulse Polarographic Determination of Trace Antimony in Standard Biological Samples after Preconcentration Using 2-Nitroso-1-naphthol-4-sulfonic Acid

A highly selective, sensitive, rapid and economical differential pulse polarographic method has been developed for the determination of trace amounts of antimony in various standard alloys and biological samples after adsorption of its 2-nitroso-1-naphthol-4-sulfonic acid - tetradecyldimethylbenzylammonium chloride on microcrystalline naphthalene in the pH range of 7.5-11.0. After filtratio...

Polarographic Determination of Traces of Gold in Blood and Serum.

A polarographic method for the determination of as little as 1 pig. of gold in blood and serum samples is described. The relative error is 2.1%. After the sample is dry ashed at 550#{176}, the gold is extracted into ether as the chloride, to separate it from interfering copper and iron. A polarogram of the gold sample is obtained in 2 M potassium hydroxide. The method should be applicable to th...
